Fiddled around with the flash file in a media player (you can skip frames, which lets you reset or swap levels), and I don't see any difference if I pick up the first mushroom twice. Either the code thought of this dickery, making each individual mushroom a separate buff, or there's only one mushroom boost.
I'm going to put my money on the latter.
